Bach1 regulates the liver specificity and transience of the Nrf2a (Montrer NFE2L2 Anticorps)-dependent induction of hmox1a and that heme mediates this regulation through Bach1 inhibition based on its level in each tissue.
The Bach1b-MafK heterodimer represses the zymogen promoters, whereas the Nrf2a-MafK heterodimer activates them.
This study showed that RANKL (Montrer TNFSF11 Anticorps) favors osteoclastogenesis via attenuation of Nrf2 (Montrer NFE2L2 Anticorps)-mediated antioxidant enzyme expression by competing with Bach1 nuclear accumulation.
this study provides new insight into the function of iron and Bach1 in erythroblast maturation by using efficacious regimens that can induce severe ID.
miR (Montrer MLXIP Anticorps)-196a, which binds to the 3'-untranslated region of the transcriptional repressor BTB and CNC homology 1 (Bach1) and regulates its expression, and subsequently leads to higher levels of lung HO-1 (Montrer HMOX1 Anticorps) mRNA.
Bach1 siRNA exerts protective effects against bleomycin-induced pulmonary fibrosis in mice.
These results suggest that Bach1 and Bach2 (Montrer BACH2 Anticorps) work in a complementary manner to maintain the normal function of the AMs (Montrer MAT1A Anticorps) and surfactant homeostasis in the lung.
By integrating the chromatin immunoprecipitation and next-generation sequencing data with a comparative analysis of gene expression in Bach1-deficient and wild type immortalized mouse embryonic fibroblasts (iMEFs), study found 35 new genes whose expression was repressed by BACH1. Known functions of some of these genes suggested a role of BACH1 in adipocyte differentiation, which was verified using iMEFs.
Bach1 deficiency reduces the severity of osteoarthritis-like changes. This may be due to maintenance of cartilage homeostasis and joint health by antioxidant effects
Bach1 suppresses angiogenesis after ischemic injury and impairs Wnt (Montrer WNT2 Anticorps)/beta-catenin (Montrer CTNNB1 Anticorps) signaling by disrupting the interaction between beta-catenin (Montrer CTNNB1 Anticorps) and TCF4 (Montrer TCF4 Anticorps) and by recruiting histone deacetylase 1 (Montrer HDAC1 Anticorps) to the promoter of TCF4 (Montrer TCF4 Anticorps)-targeted genes.
Bach1 deficiency and accompanying overexpression of HO-1 (Montrer HMOX1 Anticorps) did not influence aging or p53 (Montrer TP53 Anticorps) deficiency-driven tumorigenesis.
Our results suggest that Bach2 (Montrer BACH2 Anticorps) functions with Bach1 and EBF1 (Montrer EBF1 Anticorps) to promote B cell development by repressing myeloid genes in common lymphoid progenitor cells
The electrophilic character of quinones ensure their conjugation with Bach1, which is important for the downregulation of Bach1 and the upregulation of Nrf2 (Montrer GABPA Anticorps) signaling.
Our results suggest that the bach1-specific small interfering RNA effectively decrease CXCR4 (Montrer CXCR4 Anticorps) receptor, matrix metalloproteinase-9 (Montrer MMP9 Anticorps) expression and breast adenocarcinoma cells invasive, also increased the expression of tumor-suppressive microRNA-203 and miR (Montrer MLXIP Anticorps)-145
BACH1 down-regulation in HT29 colon cancer cells had no effect on cell growth but did inhibit cell migration by decreasing metastasis-related genes expression.
This study indicates that the expression of BACH1 could be elevated as a compensatory mechanism to decrease the globin chain imbalance as well as to reduce the oxidative stress found in hemoglobin E/beta-thalassemia.
A short while ago some studies suggested that BACH1 is involved in cancers, especially breast cancer. This factor is mentioned as a novel master regulator which adjusts several genes involved in bone metastasis of breast cancer, especially CXCR4 (Montrer CXCR4 Anticorps) and MMP1 (Montrer MMP1 Anticorps), two main genes in the enhancement of cancer cell migration and invasion to distant organs. [Review]
Bach1 suppresses cell proliferation and induces cell-cycle arrest and apoptosis.
Induction of GSH-related genes xCT (Montrer SLC7A11 Anticorps) and GCLM (Montrer GCLM Anticorps) were oxygen and Bach1-insensitive during long-term culture under 5% O2, providing the first evidence that genes related to GSH synthesis mediate protection afforded by Nrf2 (Montrer GABPA Anticorps)-Keap1 (Montrer KEAP1 Anticorps) defense pathway
heme oxygenase-1 (Montrer HMOX1 Anticorps) expression is induced by gold nanoparticles through Nrf2 (Montrer GABPA Anticorps) activation and Bach1 export in human vascular endothelial cells
Cyanidin-3-O-glucoside protects HUVECs from palmitic acid-induced injuryby modulating the balance of Nrf2 (Montrer GABPA Anticorps) versus Bach1 inside the nucleus so influencing upregulation of electrophile responsive element mediated gene expression.
sensitizer-induced up-regulation of both the endogenous HMOX1 (Montrer HMOX1 Anticorps) and the luciferase constructs under the control of the HMOX1 (Montrer HMOX1 Anticorps)-ARE or the full HMOX1 (Montrer HMOX1 Anticorps) promoter appear to be under the control of both Nrf2 (Montrer GABPA Anticorps) and Bach1.
This gene encodes a transcription factor that belongs to the cap'n'collar type of basic region leucine zipper factor family (CNC-bZip). The encoded protein contains broad complex, tramtrack, bric-a-brac/poxvirus and zinc finger (BTB/POZ) domains, which is atypical of CNC-bZip family members. These BTB/POZ domains facilitate protein-protein interactions and formation of homo- and/or hetero-oligomers. When this encoded protein forms a heterodimer with MafK, it functions as a repressor of Maf recognition element (MARE) and transcription is repressed. Multiple alternatively spliced transcript variants have been identified for this gene.
