Signalisation Notch

La voie de signalisation Notch, bien conservée, est unique en son genre car le récepteur Notch et la plupart de ses ligands respectifs (canoniquement ceux de la famille DSL ou Delta/Serrate/LAG-2) sont des protéines transmembranaires fixées à la surface de la cellule. Par conséquent, la voie de signalisation Notch se limite à l'interaction entre les cellules adjacentes. La communication entre les cellules adjacentes est primordiale, en particulier durant le début de la croissance, lorsque le destin et la fonction cellulaires doivent être encore déterminés. La voie de signalisation Notch fournit une méthode aux cellules pour indiquer leur propre identité et pour simultanément influencer le rôle et l'identité des cellules voisines par l'inhibition latérale.

La voie de signalisation Notch implique essentiellement deux cellules adjacentes, l'une exprimant un ligand de la famille DSL, et l'autre Notch (le récepteur). Quand le récepteur et le ligand interagissent, deux enzymes protéases distinctes fendent les protéines Notch en composants extracellulaires et cytosoliques. ADAM10 fend la partie extracellulaire des restes de la protéine Notch liée à son ligand respectif puis elle est endocytosée par la cellule exprimant le ligand. La γ- secrétase fend la partie cytosolique de Notch. Cette région cytosolique subit une migration vers le noyau où elle se lie au facteur de transcription CSL, le faisant passer d'un répresseur transcriptionnel à un activateur, et régulant positivement l'expression des gènes cibles de Notch.
