Plus Kits ELISA pour CHEK1 partenaires d'interaction

Human Checkpoint Kinase 1 (CHEK1) interaction partners

  1. High CHK1 expression is associated with increased radioresistance of non-small cell lung cancer.

  2. CHEK1 loss-of-function mutations have not been found in human tumors, and transgenic expression of Chek1 in mice promotes oncogene (Montrer RAB1A Kits ELISA)-induced transformation. [review]

  3. persistence of CHK1 levels in response to DNA damage in p53 (Montrer TP53 Kits ELISA)-deficient cancer cells, leads to CHK1-mediated activation of NF-kappaB (Montrer NFKB1 Kits ELISA) and induction of NF-kappaB (Montrer NFKB1 Kits ELISA)-regulated genes in cells and in associated tumor-derived microvesicles, both of which are abrogated by loss or inhibition of CHK1.

  4. Chk1's expression is controlled by p53 (Montrer TP53 Kits ELISA) and RB/E2F1 (Montrer E2F1 Kits ELISA) at the transcriptional level.

  5. High CHK1 expression correlates with urinary bladder cancer.

  6. Inappropriate activation of CDK2 (Montrer CDK2 Kits ELISA) in S phase underlies the sensitivity of a subset of cell lines to Chk1 inhibitors.

  7. In conclusion, this study exemplifies cancer-specific synthetic lethality between two proteins in the same pathway and raises the prospect of combining ATR (Montrer ANTXR1 Kits ELISA) and CHK1 inhibitors as promising cancer therapy.

  8. This study shows that Chk1 indeed maintains a closed conformation in the absence of DNA damage through an intramolecular interaction between a region (residues 31-87) at the N-terminal kinase domain and the distal C terminus. A highly conserved Leu-449 at the C terminus is important for this intramolecular interaction.

  9. Avoiding damage formation through invalidation of Mus81 (Montrer MUS81 Kits ELISA)-Eme2 (Montrer EME2 Kits ELISA) and Mre11 (Montrer MRE11A Kits ELISA), or preventing damage signaling by turning off the ATM (Montrer ATM Kits ELISA) pathway, suppresses the replication phenotypes of Chk1-deficient cells.

  10. Chk1 is a predictive biomarker of radiotherapy resistance and early local recurrence.

Xenopus laevis Checkpoint Kinase 1 (CHEK1) interaction partners

  1. The study analyzes the role of Chk1 in the replication program in sperm nuclei replicating in Xenopus egg extracts by a combination of experimental and modelling approaches.

  2. ATR-Chk1 DDR pathway appears to be dispensable for the preferential association of REV1 to MMC-damaged chromatin.

  3. Results show that Chk1 negatively regulates the Treslin-mediated loading of Cdc45 onto chromatin and thereby serves to antagonize the initiation of replication.

  4. The MRN complex, in particular the nuclease (Montrer DCLRE1C Kits ELISA) activity of Mre11 (Montrer MRE11A Kits ELISA), plays an important role in the activation of Chk1 in response to stalled replication forks.

  5. DNA polymerase kappa (Montrer POLK Kits ELISA)-dependent DNA synthesis at stalled replication forks is important for CHK1 activation.

  6. APE2 (Montrer APEX2 Kits ELISA) associates with Chk1; a serine residue (S86) in the Chk1-binding motif of APE2 (Montrer APEX2 Kits ELISA) is essential for Chk1 phosphorylation, indicating a Claspin (Montrer CLSPN Kits ELISA)-like but distinct role for APE2 (Montrer APEX2 Kits ELISA) in ATR (Montrer ATR Kits ELISA)-Chk1 signaling.

  7. Data show that the death pathway is independent of ERK (Montrer MAPK1 Kits ELISA) but relies on activating Bad phosphorylation through the control of both kinases Cdk1 (Montrer CDK1 Kits ELISA) and JNK (Montrer MAPK8 Kits ELISA).

  8. Findings reveal that XH2AX has a specific role in anterior neural formation of Xenopus, which is mediated through phosphorylation of XH2AX at Thr (Montrer TRH Kits ELISA)(16) by Chk1.

  9. mechanism of Chk1 activation at the DNA replication checkpoint

  10. Cdc25A (Montrer CDC25A Kits ELISA) is essential for the Chk1-induced cell cycle arrest and the DNA replication checkpoint in early embryos

Mouse (Murine) Checkpoint Kinase 1 (CHEK1) interaction partners

  1. RAD9 (Montrer RAD9A Kits ELISA) has a prominent role in the ATR (Montrer ATR Kits ELISA)-Chk1 pathway that is necessary for successful formation of the damage-sensing complex and DNA damage checkpoint signaling.

  2. Acute inactivation of E4F1 (Montrer E4F1 Kits ELISA) in these cells results in CHK1-dependent checkpoint deficiency and multiple mitochondrial dysfunctions that lead to increased ROS (Montrer ROS1 Kits ELISA) production, energy stress, and inhibition of de novo pyrimidine synthesis

  3. Altogether, our results classify E4F1 (Montrer E4F1 Kits ELISA) as a master regulator of CHK1 activity that ensures high fidelity of DNA replication, thus safeguarding genome stability.

  4. Heterozygous loss of Chk1 in a Wnt (Montrer WNT2 Kits ELISA)-driven background resulted in an increase in DNA damage and apoptosis and accelerated both tumour development and progression.

  5. PAR, supplied by PARP1, interacts with Chk1 via a novel PAR-binding regulatory (PbR) motif in Chk1, independent of ATR and its activity

  6. ATM (Montrer ATM Kits ELISA) (pSer-1981)-Chk1 (pSer-345) cascade might have mediated G2/M cell cycle arrest to allowed time to facilitate sperm-derived DNA damage repair in mouse zygotes fertilized with oxygen-stressed sperm.

  7. High level of phospho-Chk1 is associated with skin tumors.

  8. Chk1 is essential for the developmental formation of murine epidermal melanocytes but hemizygosity has little permanent developmental consequence in this cell type

  9. activated Akt1 (Montrer AKT1 Kits ELISA) in Brca1 (Montrer BRCA1 Kits ELISA)-deficient cells impairs Chk1 nuclear localization and subsequently disrupts interaction of Chk1 and Rad51 (Montrer RAD51 Kits ELISA) leading to homologous recombination defects.

  10. Our results uncover a novel p53 (Montrer TP53 Kits ELISA)-independent function for p21 in protecting cells from the lethal effects of DNA damage followed by Chk1 inhibition

Cow (Bovine) Checkpoint Kinase 1 (CHEK1) interaction partners

  1. The Chk1 directly phosphorylates eNOS (Montrer NOS3 Kits ELISA) Ser (Montrer SIGLEC1 Kits ELISA)(1179) in response to UV irradiation, which is dependent on Hsp90 (Montrer HSP90 Kits ELISA) interaction.

CHEK1 profil antigène

Antigen Summary

The protein encoded by this gene belongs to the Ser/Thr protein kinase family. It is required for checkpoint mediated cell cycle arrest in response to DNA damage or the presence of unreplicated DNA. This protein acts to integrate signals from ATM and ATR, two cell cycle proteins involved in DNA damage responses, that also associate with chromatin in meiotic prophase I. Phosphorylation of CDC25A protein phosphatase by this protein is required for cells to delay cell cycle progression in response to double-strand DNA breaks. Several alternatively spliced transcript variants have been found for this gene.
