Signalisation PI3K-Akt

La transduction de signal PI3K/Akt a un rôle charnière dans l’homéostasie cellulaire, le développement neurologique, le métabolisme et d’autres processus. Elle régule différents aspects du développement cellulaire comme l’apoptose, la progression du cycle cellulaire et la différenciation cellulaire.

Le régulateur principal du signal PI3K/Akt est Akt/PKB, une famille de trois protéines kinases sérine/thréonine apparentées les unes aux autres. Akt1/PKB alpha joue un rôle important dans la prolifération et le métabolisme cellulaires. Akt2/PKB beta a une grande importance dans le métabolisme du glucose. Akt3/PKB gamma est le membre le moins caractérisé du groupe Akt mais il est surtout exprimé dans le cerveau, où l’on pense qu’il régule la biogenèse mitochondriale.

Les mutations d’Akt contribuent souvent à la dérégulation de ces processus et à l’apparition de différentes formes de cancer. L’hyper-activation d’Akt conduit également à une augmentation de la glycolyse, ce qui favorise la transformation métabolique des cellules cancéreuses. Akt a été utilisé dans le développement de résistance aux médicaments contre le cancer et fait donc l’objet de recherches précliniques et cliniques. Cependant, l’activation d’Akt à elles-seules ne sont d’ordinaire pas suffisantes and d’autres mutations sont généralement nécessaires pour créer le phénotype du cancer.

La perte de fonction des régulateurs principaux Akt PI3K et PTEN peut conduire à l’hyper-activation d’Akt. Les PI3K sont une famille d’enzymes qui provoquent la phosphorylation de PtdIns (phosphatidylinositol) pour générer du phosphatidylinositol 3,4,5-trisphosphate (PIP3). Sur activation par le récepteur à activité tyrosine kinase, PI3K provoque la phosphorylation de PIP2 vers PIP3, ce qui active ensuite la transduction de signal Akt. Le PTEN agit comme un régulateur négatif de la transduction de signal par déphosphorylation de PIP3 vers PIP2.
