FBXL21 plays a dual role: protecting CRY (Montrer CRY2 Protéines) from FBXL3 (Montrer FBXL3 Protéines) degradation in the nucleus and promoting CRY (Montrer CRY2 Protéines) degradation within the cytoplasm.
Study shows that FBXL21 ubiquitinates CRY1 (Montrer CRY1 Protéines) and CRY2 (Montrer CRY2 Protéines) and that, surprisingly, FBXL21 stabilizes CRYs. FBXL21 localizes predominantly in the cytosol, whereas FBXL3 (Montrer FBXL3 Protéines) is present in the nucleus.
Using both family and case-control samples, consistent associations in and around FBXL21 gene suggested that FBXL21 may be associated with schizophrenia in the Irish samples.
This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class and, in addition to an F-box, contains 6 tandem leucine-rich repeats. The amino acid sequence of this protein is highly similar to that of f-box and leucine-rich repeat protein 3A. Comparisons of this gene to orthologous sequences suggest that it may be a pseudogene, and may no longer express a functional protein.
