PA28, also called 11S regulator, was identified as a proteasome activator that strongly increases the maximal velocity of the hydrolytic reaction and decreases the concentration of substrate required for cleavage by purified 20S proteasomes. It is known that major histocompatibility complex (MHC) class I ligands are mainly produced by the proteasome. PA28 also plays an important role in the processing of MHC class I ligands. However, isolated PA28 does not perform enzymatic activity by itself. PA28 is composed of two homologous subunits, PA28α (30 kDa) and PA28ß. These two subunits assemble into a heterohexamer (α3ß3) or a heteroheptomer (α3ß4 or mixture of α3ß4 and α4ß3).
