Immunohistochemistry: This antibody stained formalin-fixed paraffin-embedded sectionsof human pancreas infiltrating ductal adenocarcinoma tissue. The recommendedconcentration is 1.0-2.0 μg/mL with an overnight incubation at 4 °C. An HRP-labeledpolymer detecttion system was used with an alcohol-soluble AEC chromogen. Optimalresults for these conditions were achieved with heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer enzyme induced antigen retrievalwith proteinase K at roomtemperature.
Optimal conditions may vary. Tissue samples were provided by theCooperative Human Tissue Network which is funded by the National Cancer Institute. Indirect ELISA: To detect Human MIA (using 100 μL/well antibody solution) a concentrationof 0.5-2.0 μg/mL of this antibody is required. In conjunction with compatible secondaryreagents, it allows the detection of at least 0.2-0.4 ng/well of recombinant Human MIA. Sandwich ELISA: To detect Human MIA (using 100 μL/well antibody solution) aconcentration of 0.5-2.0 μg/mL of this antibody is required. In conjunction withBiotinylated Anti-Human MIA (ABIN181670 or ABIN181669) as a Detection antibody, itallows the detection of at least 0.2-0.4 ng/well of recombinant Human MIA. Western blot: To detect Human MIA this antibody can be used at a concentration of 0.1-0.2 -

- Melanoma Inhibitory Activity (MIA) was originally identified as an inhibitor of the in vitro growth of malignant melanoma cells. It was the first discovered member of a family of secreted cytokines termed the MIA/OTOR family. The four known members of this family, Melanoma Inhibitory Activity, MIA2, OTOR and TANGO each contain a Src homology-3 (SH3)-like domain. Melanoma Inhibitory Activity is an autocrine growth regulatory protein secreted from chondrocytes and malignant melanoma cells that promotes melanoma metastasis by binding competitively to fibronectin and laminin in a manner that results in melanoma cell detachment from the extracellular matrix in vivo. Elevated levels of Melanoma Inhibitory Activity may represent a clinically useful marker for diagnosis of melanoma metastasis as well as a potential marker for rheumatoid arthritis.Synonyms: Melanoma inhibitory activity, Melanoma-derived growth regulatory protein
