Description: Complement fragment C5a is a pro-inflammatory polypeptide produced during activation of the complement cascade of serum proteins in response to foreign surfaces such as microorganisms and tissue damaged by physical or chemical injury. C5a binds to at least two seven-transmembrane domain receptors, C5aR (C5R1, CD88) and C5L2 (gpr77).Expression of C5L2 (C5a Anaphylatoxin Receptor 2) can be found in various tissues and cells such as astrocytes, neutrophils/macrophages, mast cells, immature dendritic cells, as well as in the brain, lung, heart, kidney, liver, ovary or testis.The C5L2 receptor is a G-protein coupled receptor clustered with other chemoattractant receptors, such as type-II angiotensin-II receptor, bradykinin receptors, the formyl peptide receptor family, ChemR23 and several orphan GPCRs. A small number of single-nucleotide polymorphisms (SNP) have been found in the receptor gene. C5L2 has two synonymous SNPs at 614G/A and 860C, however there are no known associations between these SNPs and human disease.Although C5L2 has the conventional structure of a GPCR, studies have shown that C5L2 does not couple to G proteins owing to the lack of a highly conserved Asp-Arg-Tyr motif, found in the third TM domain1.
