Description: The Mas-related G protein-coupled receptors (MRGPRs), formerly called Mas-related genes (MRGs) includes nine members: MrgprA, B, C, D, E, F, G, H, and the primate-specific MRGPRX1,2.MRGPRE consist of only one receptor per species and is conserved in rodents and primates2. MRGPRE structure contains a Cys-Cys bridge from transmembrane domain (TM) 3, an "ionic-lock" known to inhibit activation and activity in some class A GPCRs1,2.Human and mouse MRGPRE is highly expressed in neurons of the brain and mast cells. Transcripts are also found in medium and large diameter neurons of human dorsal root ganglia (DRG) sections and in other areas of the central nervous system, including cerebral cortex, hippocampus, spinal cord, and cerebellum. MRGPRE was also found in human, mouse, and rat placenta tissue and in enteric neurons1,2.Mouse lacking the MRGPRE protein showed decreased nociceptive behavior and significant deficits in the induction of mechanical allodynia after chronic constriction injury2.
