- CD8, known as Cluster of Differentiation 8, forms a disulphide-linked heterodimer comprising unrelated α and β subunits. Each subunit, characterized as a glycoprotein, is structured with a single extracellular Ig-like domain, a polypeptide linker, a transmembrane component, and a concise cytoplasmic tail. Within T cells, CD8 serves as the co-receptor alongside the T cell receptor (TCR). These distinct structures collaboratively identify the Antigen-Major Histocompatibility Complex (MHC). Specifically, the interaction occurs between the Ig-like domain of CD8α and the α3-domain of the MHC class I molecule. CD8 plays a pivotal role in ensuring the specificity of the TCR-antigen interaction, extending the duration of contact between the T cell and the antigen-presenting cell. Moreover, the α chain of CD8 recruits the tyrosine kinase Lck, an indispensable component for initiating T cell activation .
