Background_Plain: ING5 (Inhibitor of Growth 5) possesses different structural and functional elements, including a PHD (plant homeodomain), an NCR (novel conserved region), a LZL (leucine zipper like) finger, and two NLS (nuclear localization sequence) motifs. ING proteins bind to methylated core histones through their PHD motif, notably ING5 reads histone H3 when di- or trimethylated at lysine 4 (H3K4me2/3). ING5 is a subunit of two distinct histone acetyltransferase (HAT) complexes with HBO1 or MOZ/MORF as the two catalytic subunits. These complexes contain two different alternative scaffolding proteins, i.e. a BRPF protein or a JADE protein, that determine their specificity towards different core histones. ING complexes appear to participate in transcription, for example by interacting with transcriptional regulators such as the tumor suppressor p53 and through the ability of ING5 to recognize H3K4me3, a mark associated with open chromatin and transcribed genes.
