Sujet: Adipocytes express a variety of proteins that function in the homeostatic control of glucose and lipid metabolism. Insulin regulates the translocation and secretion of many of these adipocyte proteins in response to homeostatic changes in energy balance. Both adipsin and adipocyte complement-related protein of 30 kDa (Acrp30) are secretory proteins whose secretion from adipocytes is enhanced by insulin stimulation. Acrp30 contains N-terminal collagen repeats and a globular C-terminal domain. It is homologous to collagen type VIII and X and to complement factor C1q. Also, it forms homo-oligomeric structures of 90 and 300 kDa. The expression of Acrp30 increases 100 fold during adipocyte differentiation and it is commonly found in serum. Insulin stimulation of 3T3-L1 fibroblasts leads to a 4-fold increase in Acrp30 secretion. This secretion is regulated by PI3-kinase activity and may involve a secretory pathway that is distinct from other insulin-regulated secretory pathways, such as that involved with the translocation of glucose transporters and transferrin receptors. Thus, Acrp30 is an adipocyte secretory protein that functions in the regulation of energy homeostasis.
