- The microtiter plate provided in this kit has been pre-coated with an antibody specific to MAL. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for MAL and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain MAL, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of MAL in the samples is then determined by comparing the O.D. of the samples to the standard curve.

- Maltase (acid maltase, alpha-1,4-glucosidase) is an enzyme produced by the cells lining the small intestine that breaks down the disaccharide maltose. It comes under the enzyme category carbohydrase (which is a subcategory of hydrolase). Maltase is secreted by the surface cells of the villi, which are thin projections on the mucosa. These are found throughout the small intestine, but differ in shape in the duodenum and ileum sections. It must also be located in other places, like the heart, if a deficiency of it causes Pompe's Disease (which results in cardiomegaly).
