Indications d'application: Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.

Protocole: ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. An antibody has been precoated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection antibody specific for target is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the target amount in sample captured in plate.

Background: JAM3 belongs to the junctional adhesion molecule protein family and functions as a receptor for another member of this family. This gene is mapped to 9, 9 A4. JAM3 plays a part in cell-cell adhesion. The soluble form of JAM3 is a mediator of angiogenesis. JAM3 is a counter-receptor for ITGAM, mediating leukocyteplatelet interactions and is involved in the regulation of transepithelial migration of polymorphonuclear neutrophils (PMN). A mutation in an intron of the JAM3 gene is linked with hemorrhagic destruction of the brain, subependymal calcification, and congenital cataracts.
