Serum IgA1 is the predominant subclass of Immunoglobulin A in human circulation, comprising approximately 90% of total serum IgA. It eatures a distinctive hinge region with multiple O-linked glycans, distinguishing it structurally from IgA2. IgA1 serves critical immune functions by neutralizing pathogens, facilitating immune complex clearance, and mediating antibody-dependent cell-mediated cytotoxicity through FcαRI receptors on neutrophils and monocytes.
Aberrant glycosylation of IgA1 plays a central role in IgA nephropathy pathogenesis, where galactose-deficient IgA1 forms immune complexes that deposit in glomeruli, triggering kidney inflammation and damage. Additionally, IgA1 abnormalities are implicated in IgA vasculitis, celiac disease, and certain autoimmune conditions.
