    Protein Background: PARP7(Protein mono-ADP-ribosyltransferase TIPARP), also known as TiPARP, is a mono ADPribosyltransferase belonging to the Poly-ADP-ribose polymerase (PARP) family. Unlike some PARPs involved in DNA repair, PARP7's primary functions are linked to regulating gene expression and the immune response, particularly the type I interferon signaling pathway. It is induced by the transcription factor AHR (aryl hydrocarbon receptor) and acts as a negative feedback regulator. PARP7 modifies itself and other target proteins with a single ADP-ribose unit (MARylation), which influences their activity and stability. By MARylating key transcription factors like AHR and IRF3/7, it dampens inflammatory and interferon responses, thereby maintaining immune homeostasis. Its role as a checkpoint in anti-tumor immunity has made it a emerging target for cancer immunotherapy, where its inhibition is sought to enhance the body's innate immune attack against cancer cells.
