    anti-TBEV IgG (CSF) assay is a solid-phase immunoanalytical test. The polystyrene strips are coated with specific antigen which bear immunodominant epitopes of TBEV. Anti-TBEV antibodies in samples bind to the immobilized antigens. The antibodies that do not bind are washed away and those that formed complexes with the antigens are later on recognised by animal anti-human IgG antibodies labelled with horseradish peroxidase. The presence of labelled antibodies is revealed by an enzymatic reaction with a chromogenic substrate. Negative samples do not react and the mild change in colour, if present, may be attributed to the reaction background. Intrathecal antibody testing provides information on the anti-TBEV antibody response in the central nervous system. For this assay, it is necessary to quantify the concentration of TBEV-specific IgG antibodies in paired serum / plasma and cerebrospinal fluid samples taken from the patient at the same time. Accurate quantification of antibodies can only be performed according to the linear part of the calibration curve. Therefore, it is recommended to test the serum in two dilutions (see Chapter 6 Reagent preparation). The total IgG and albumin concentrations in both samples must be known for the determination. The calculation of intrathecal synthesis of specific antibodies is performed using the Reiber equation (see Chapter 8.4 Intrathecal synthesis of specific IgG antibodies).
