  • Indications d'application
    ELISA: In a sandwich ELISA (assuming 100 μL/well), a concentration of 3-4 μg/mL of thisantibody will detect at least 0.1 ng/well of recombinant murine IL-12 when used withbiotinylated antigen affinity purified anti-murine IL-12 as the detection antibody. Neutralisation: To yield one-half maximal inhibition (ND50) of the biological activity ofmurine IL-12 (2.0 ng/mL), a concentration of 0.16-0.24 μg/mL of this antibody is required. Western Blot: To detect mIL-12 by Western Blot analysis this antibody can be used at aconcentration of 2.0-4.0 μg/mL. Used in conjunction with compatible secondary reagentsthe detection limit for recombinant mIL-12 is 1.0-2.0 ng/lane, under reducing conditionsand 0.25-0.50 ng/lane, under non-reducing conditions.
    Other applications not tested.
    Optimal dilutions are dependent on conditions and should be determined by the user.

    Interleukin 12 (IL12) can act as a growth factor for activated T and NK cells, enhance the lytic activity of NK/lymphokine activated killer cells, and stimulate the production of IFN gamma by resting PBMC. IL12 induces IFN gamma, TNF alpha production in T and NK cells, costimulation of PBL proliferation, proliferation/differentiation of TH1 T lymphocytes. It is downregulated by IL10, homodimeric p40 antagonistic to functional p70 heterodimer.
