- SIRT6 (SIR2-like protein 6) is a member of the yeast silent information regulator 2 (SIR2) family and possesses both strong ADP-ribosyltransferase activity as well as protein deacetylase activity which is dependent on NAD(+). SIRT6 exerts deacetylase activity towards histone H3K9Ac and H3K56Ac, and alters histone H3 acetylation in telomeric chromatin during cell cycle's S-phase. It deacetylates histone H3K9Ac at NFkB target promoters and downregulates a subset of NFkB target genes. Upon DNA damage, SIRT6 promotes DNA end resection via deacetylation of RBBP8. SIRT6 also serves as a corepressor of the transcription factor HIF1 alpha for regulating the expression of multiple glycolytic genes to control glucose homeostasis, and is required for normal IGF1 serum levels. SIRT6 play critical roles in maintaining telomere integrity, fine-tuning aging-associated gene expression programs, preventing genomic instability, and maintaining metabolic homeostasis, thus impacting on several pathways relevant for cancer, metabolism, aging, cellular senescence and apoptosis.
