Each AbFlex antibody contains a 6xHis Tag, a Biotinylation Tag for enzymatic biotin conjugation using the biotin ligase, BirA, and a sortase recognition motif (LPXTG) to attach a variety of labels directly to the antibody including fluorophores, enzymatic substrates (HRP, AP), peptides, drugs as well as solid supports.

Indications d'application
- WB*: 0.1 - 1 μg/mL *Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western blot. This recombinant antibody has been generated using defined DNA sequences to achieve high specificity and high reproducibility. The unique advantages of this antibody are its flexible labeling and purification options as it contains a sortase recognition motif (LPXTG) to covalently add fluorophores, enzymatic substrates (HRP, AP...etc), peptides, DNA, drugs or other labels to the antibody in a directed and reproducible manner. Every antibody also contains a 6xHis Tag, which can be used with nickel-based purification systems, and a Biotinylation Tag sequence for enzymatic biotin conjugation using the biotin ligase, BirA.

- AbFlex Nucleolin antibody was expressed as full-length IgG with mouse immunoglobulin heavy and light chains (IgG2a isotype) in mammalian 293 cells. Nucleolin (NCL, C23) is an abundant and ubiquitously expressed protein that is a major component of the nucleolus. Nucleolin seems to be overexpressed in highly proliferative cells and is involved in many aspects of gene expression: chromatin remodeling, DNA recombination and replication, RNA transcription by RNA polymerase I and II (RNA pol II), rRNA processing, mRNA stabilization, cytokinesis and apoptosis. Its role as a histone chaperone has recently been identified. It is involved in the synthesis and maturation of ribosomes, and is located mainly in dense fibrillar regions of the nucleolus.
