- Background: FBW7 (F-box and WD repeat domain containing 7) is a member of the F-box family of proteins. The F-box is a protein-protein interaction motif first identified in cyclin F. F-box proteins associate with SCFs (Skp1-cullin-F-box protein ligase complexes) which are E3 ligases that ubiquitinate proteins and target them for proteasomal degradation. FBW7 has been found to bind and specifically target cyclin E and the Notch intracellular domain (NICD) for ubiquitination. Defects in FBW7 may contribute to tumorigenesis.
