- Background: Bub1 is cell cycle checkpoint kinase and component of the spindle checkpoint machinery. Bub1 and other spindle checkpoint proteins ensure that segregation does not proceed until all replicated chromatids are associated with a functional bipolar spindle. Bub1 functions to target Bub1-related protein (BubR1), Mad2, and polo-like kinase to kinetochores. It also functions to phosphorylate Cdc20 and inhibit ubiquitination of Pds1/securin by the anaphase-promoting complex (APC).
