    Description: Synaptojanin 1 (SYNJ1) is a phosphoinositide phosphatase primarily found in nerve terminals, where it plays a critical role in regulating synaptic vesicle recycling and neurotransmission, membrane trafficking and actin cytoskeleton dynamics. It belongs to the synaptojanin protein family, which includes other isoforms (SYNJ2 and SYNJ2BP) that also participate in cellular membrane dynamics and endocytic processes.1 Synaptojanin 1 is unique among phosphoinositide phosphatases in that it contains two catalytic domains that act on different substrates within the phosphoinositide signaling pathway. The SAC1 domain hydrolyzes phosphatidylinositol 3-phosphate (PI3P) and phosphatidylinositol 4-phosphate (PI4P), while the 5-phosphatase domain dephosphorylates phosphatidylinositol 4,5-bisphosphate (PI(4,5)P₂) to PI4P and phosphatidylinositol 3,4,5-trisphosphate (PI(3,4,5)P₃) to PI(3,4)P₂. 1,2 The coexistence of SAC1 and 5-phosphatase domains enables Synaptojanin 1 to orchestrate multiple steps in membrane trafficking across different subcellular compartments including vesicle uncoating, membrane trafficking, receptor recycling, and autophagy, processes fundamental to neuronal health, synaptic transmission, and plasticity.1,2,3 Based on its key function Synaptojanin 1 emerged as a key player in both short-term synaptic dynamics and long-term synaptic remodeling. In fact, Synaptojanin 1 dysfunction has been implicated in several neurodevelopmental and neurodegenerative disorders.3 Mutations in Synaptojanin 1 have been identified to result in comorbidities of early-onset Parkinsonism and epilepsy. The condition is now classified as PARK20, an autosomal recessive, juvenile-onset parkinsonism that presents with bradykinesia, dystonia, seizures, and cognitive impairment.3,4 Elevated levels of Synaptojanin 1, have been found in both human Alzheimer's disease (AD) brains, as well as mouse AD models and are correlated with cognitive decline linked to age.3,5
