    Many mitochondrial proteins are encoded by nuclear genes and synthesized in the cytosol as pre-proteins. These pre-proteins are targeted to the mitochondria where they are translocated across the outer and inner membranes of the mitochondria. At the outer membrane of the mitochondria, this process requires a multiprotein complex called the Translocase of the Outer Membrane of the mitochondria (TOM). The TOM complex includes receptor components, Tom20, 22, 37, and 70, as well as a multisubunit general import pore complex of 400 kDa. Tom20 contains a hydrophobic N-terminal region that anchors it to the outer membrane of the mitochondria, and a cytosolic region that contains a domain that facilitates binding to the basic amphiphilic targeting sequences of pre-proteins and a tetratricopeptide repeat (TPR) motif involved in interactions with Tom70. In yeast, Tom20 and Tom22 mediate the import of all pre-proteins known to use the general import machinery of the mitochondria. Thus, Tom20 is an important receptor for many mitochondrial pre-proteins that use the outer mitochondrial translocation complex.
