Indications d'application: Before using Kit, spin tubes and bring down all components to bottom of tube. Duplicate well assay was recommended for both standard and sample testing.

Protocole: human IL-17RA ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for IL-17RA has been precoated onto 96-well plates. Standards (NSO, L33- W320) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for IL-17RA is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human IL-17RA amount of sample captured in plate.

Procédure de l'essai:

Aliquot 0.1 mL per well of the 2000pg/mL,1000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, 62.5pg/mL, 31.2pg/mL human IL-17RA standard solutions into the precoated 96-well plate. Add 0.1 mL of the sample diluent buffer into the control well (Zero well). Add 0.1 mL of each properly diluted sample of human cell culture supernates, serum and plasma (heparin, EDTA) to each empty well. See "Sample Dilution Guideline" above for details. It is recommended that each human IL-17RA standard solution and each sample be measured in duplicate.

Background: Interleukin 17 receptor A, also known as IL17RA and CDw217 (cluster of differentiation w217), is a human gene. The IL17R gene is mapped to 22q11.22-q11.23 by radiation hybrid analysis. Interleukin 17A (IL17A) is a proinflammatory cytokine secreted by activated T-lymphocytes. It is a potent inducer of the maturation of CD34-positive hematopoietic precursors into neutrophils. The protein encoded by this gene is a ubiquitous type I membrane glycoprotein that binds with low affinity to interleukin 17A. Interleukin 17A and its receptor play a pathogenic role in many inflammatory and autoimmune diseases such as rheumatoid arthritis. Like other cytokine receptors, this receptor likely has a multimeric structure. Also, it is showed that mouse Il17r bound HSV-13 and mouse Il17. Recombinant soluble Il17r inhibited mitogen-induced T-cell proliferation and Il2 production.
