Usage: Suitable for use in Direct ELISA, Western Blot, Flow Cytometry and Immunohistochemistry. By Flow Cytometry and Immunohistochemistry, this antibody detects mouse Frizzled-3 on differentiated embryoid bodies derived from the mouse embryonic cell line, D3. Flow Cytometry: Dilute this antibody to 5-10 μg/mL and add 10 mL of the diluted solution to 1-2.5x10^5 cells in a total reaction volume not exceeding 200 μL. The binding of unlabeled monoclonal antibodies may be visualized by adding 10 μL of a 25 μg/mL stock solution of a secondary developing reagent such as goat anti-rat IgG conjugated to a fluorochrome. Immunohistochemistry: A biotin conjugate of this antibody at a concentration of 8 μg/mL was used to detect Frizzled-3 in frozen mouse embryo sections. Western Blot: Use at 1-2 μg/mL with the appropriate secondary reagents to detect mouse Frizzled-3. Using a colorimetric detection system, the detection limit for rmFrizzled-3 is ~1 ng/lane under non-reducing and reducing conditions.
