Description: Tetraspanins (Tspans) are a family of four-span transmembrane proteins, known as plasma membrane "master organizers." They form Tspan-enriched microdomains (TEMs or TERMs) through lateral association with one another and other membrane proteins.1Tetraspanins are characterized by four transmembrane domains, two extracellular loops: one small extracellular loop (SEL or EC1) and one larger extracellular loop (LEL or EC2), a short intracellular loop, and two short intracellular tails.1Teatraspanin 7 (TSPAN7) is highly expressed in the brain and was first described as a candidate gene for X-linked intellectual disability, probably through the modulation of surface expression of AMPA and dopamine receptors.2Tetraspanin 7 also has a key role in controlling pancreatic β-cell Ca2+ handling and defining the set-point of insulin secretion through modulation of L-type Cav1.2 and Cav1.3 channels. Furthermore, it has been identified as a target for autoantibodies in Type I diabetes. 3In addition, Tetraspanin 7 is involved in the regulation of HIV virus entry into dendritic cells and several types of cancer progression. 2
