    Specificité

The antibody was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific phosphopeptide. The antibody against non-phosphopeptide was removed by chromatogramphy using non-phosphopeptide corresponding to the phosphorylation site. ASK1 (phospho-Ser83) antibody detects endogenous levels of ASK1 only when phosphorylated at serine 83.

    Purification

    Immunoaffinity chromatography using epitope-specific phosphopeptide.

    Immunogène

The antiserum was produced against synthesized phosphopeptide derived from human ASK1 around the phosphorylation site of serine 83 (G-S-Sp-V-G).

    Sujet

Mitogen activated protein (MAP) kinase cascades are activated in response to various extracellular stimuli including cytokines, growth factors and environmental stresses. A novel MAP kinase kinase kinase (MAPKKK) was recently identified and designated ASK1 (for apoptosis signal-regulating kinase 1). ASK1 activates two different subgroups of MAPKK, MKK4 and MKK6, which in turn activate c-Jun N terminal kinase (JNK) and p38 MAP kinase, respectively. ASK1 is activated by TNFR and Fas through the interaction with members of the TRAF family and Fas associated protein Daxx. Overexpression of ASK1 induces apoptotic cell death, and a catalytically inactive form of ASK1 inhibits TNF alpha-induced apoptosis. ASK1 is expressed in variety of human and mouse tissues.Synonyms: Apoptosis signal-regulating kinase 1, MAP3K5, MAPK/ERK kinase kinase 5, MAPKKK5, Mitogen-activated protein kinase kinase kinase 5
