Indications d'application

Suitable for Western Blot (0.5 μg/mL for HRPO/ECL detection)Recommended buffer: Casein/Tween 20 based blocking and blot incubation buffer. Use SW480 pervanadate-treated as positive control (see Protocols below). ELISA: 0.1 μg/mL.
Other applications not tested.
Optimal dilutions are dependent on conditions and should be determined by the user. -
Protocole
- Contains Positive Control (pervanadate treated SW 480 cells)Format: Lyophilized cell lysate from SW480 cells. Serum starved cells were treated for15min with pervanadate. Reconstitution: Add 200 µl H2O. After complete solubilization add 200 µl 2x SDS-PAGEsample buffer, mix and incubate at 90°C for 5 min. Application: The positive control cell lysate is recommended for immunoblot applications. 20 µl of positive control cell lysate correspond to ca. 20. 000 cells. Use 20 µl/lane (mini gel) for HRP/ECL detection of the target proteins. Please note: The lyophilized cell lysates contain SDS and are not recommended forapplications with native proteins such as immunoprecipitation. Aliquot and store frozen. Avoid repeated freeze/thaw cycles.

- MEK (MAP Kinase Kinase) phosphorylates the MAP Kinase 1 and 2 on both threonine and tyrosine residues of the activation loop motif TEY. MEK1 and MEK2 are activated by phosphorylation of two serine residues (Ser 218/222 in MEK1 and Ser 222/226 in MEK2).
