ELISA: 1/10,000-1/50,000. Western blot: 1/500-1/2,000 (Expect a band approximately 69 kDa in size corresponding toFANCG in the appropriate human tissue). FANCG (also called Protein FACG or DNA-repair protein XRCC9) is involved in DNA repair, perhaps specifically with post-replication repair or a cell cycle checkpoint function. FANCG may also be implicated in interstrand DNA cross-link repair and in the maintenance of normal chromosome stability. This protein may also function as a tumor suppressor gene. FANCG belongs to the multi-subunit Fanconi Anemia (FA) complex composed of FANCA, FANCB, FANCC, FANCE, FANCF, FANCG, FANCL/PHF9 and FANCM. FANCG contains a 5-prime GC-rich untranslated region characteristic of housekeeping genes. The putative 622-amino acid protein has a leucine-zipper motif at its N-terminus. FANCG is mainly found within the nucleus although some protein is localized in the cytoplasm. This protein is highly expressed in testis and thymus and is also found in lymphoblasts. Fanconi anemia is an autosomal recessive disorder with diverse clinical symptoms, including developmental anomalies, bone marrow failure, and early occurrence of malignancies caused by defects in the FANCG gene. At the cellular level it is associated with hypersensitivity to DNAdamaging agents, chromosomal instability (increased chromosome breakage), and defective DNA repair.Synonyms: DNA repair protein XRCC9, FACG, Fanconi anemia group G protein
