- Background: Rad9 is a cell-cycle checkpoint protein with 3'-5' exonuclease activity and is part of the 9-1-1 complex. The 9-1-1 complex, also called the checkpoint clamp, is a trimeric complex that includes Rad9, Hus1, and Rad1. This complex is recruited by the RAD17/RFC clamp loader to DNA lesions where it aids in the activation of Chk2 by ATM and ATRIP and the subsequent triggering of the checkpoint signaling cascade. In addition to its role in sensing DNA damage, Rad9 also functions in the base excision repair process of DNA repair.
