- Background: JAK2 (Janus kinase 2) was originally cloned via screening of mouse cDNA libraries with PCR generated probes representing highly conserved elements in the protein tyrosine kinase (PTK) family. JAK2, as well as JAK, was named after the Roman god, Janus, the god of gates or doors, and who is depicted as having two faces which face opposite directions. JAK kinases bear two nearly identical phosphate transferring domains and function to transduce signals from type I and type II cytokine receptors for the activation of STAT (signal transducers and activators of transcription) transcription factors.
