The IL-21 receptor (IL-21R) is structurally related to IL-2R and IL-15R. In addition, these receptors, together with IL-4R, IL-7R, and IL-9R, share a common gamma -chain. IL-21R appears in T, natural killer (NK), B, and dendritic cells (DC), and it plays an important role in the regulation and function of these cells. IL-21 has, however, an effect on NK cells depending on their activation state. It enhances the proliferation and cytotoxic activity of activated NK cells, but in these cells, IL-21 also promotes apoptosis and thus limits NK cell response. In contrast, IL-21 inhibits IL-15-induced proliferation and maturation of NK cell precursors
