Peptide ELISA: antibody detection limit dilution 1: 32000. Western blot: Preliminary experiments gave an approx 50 kDa band in Human Testis andOvary lysates after 0.3 μg/mL antibody staining. Please note that currently we cannot findan explanation in the literature for the band we observe given the calculated size of11.0 kDa according to isoform a (also called HMG-I, NP_665906.1, NP_665908.1,NP_665911.1) and 10.0 kDa according to isoform b( (also called HMG-Y, NP_002122.1,NP_665909.1, NP_665910.1, NP_665912.1). The 50 kDa band was successfully blocked byincubation with the immunizing peptide. We would appreciate any feedback from peoplein the field - have any results been reported with other antibodies/lysates? Have anyfurther splice variants/modified forms been reported?Immunohistochemistry: In paraffin embedded Human Placenta shows strong nuclearstaining. Recommended concentration, 4-6 μg/mL.

HMGA1 (HMGI/Y) is a non-histone protein involved in many cellular processes, including regulation of inducible gene transcription, integration of retroviruses into chromosomes, and the metastatic progression of cancer cells. There are two splice isoforms of HMGA1 - the full length HMGA1a/HMGI and the shortened form HMGA1b/HMGY. The encoded protein preferentially binds to the minor groove of A+T-rich regions in double-stranded DNA. It has little secondary structure in solution but assumes distinct conformations when bound to substrates such as DNA or other proteins. HMGA1 is frequently acetylated and is found in the nucleus.
