  • Indications d'application
    The 53-6.7 antibody has been reported to block antigen presentation via MHC class I and inhibit T cell responses to IL-2. This antibody has also been used for depletion of CD8a+ cells.It is recommended for use in flow cytometry. This reagent is effective for direct immunofluorescence staining of mouse tissue for flow cytometric analysis usingless than or equal to 1 µg/10^6 cells.

    Préparation des réactifs
    1. Transfer the sample of blood to a 12 x 75 mm polystyrene test tube (10^6 cells). 2. Add anti-mouse CD8a FITC and mix gently with a vortex mixer. The optimal volume should be determined by the individual laboratory. 3. The recommended negative control is a non-reactive FITC-conjugated antibody of the same isotype. 4. Incubate in the dark at room temperature at 4°C for 30 minutes or at room temperature (20-25 °C) for 15 minutes. 5. Add 1,5 mL of Lysing Solution to each sample and mix gently with a vortex mixer. Incubate for 10 minutes at room temperature in the dark. 6. Centrifuge at 1000 x g for 5 minutes. Gently aspirate the supernatant and discard it leaving approximately 50 µl of fluid. 7. Add 2 mL 0.01 mol/L PBS (It betters that it containing 2% bovine serum albumin) and resuspend the cells by using a vortex mixer. 8. Centrifuge at 1000 x g for 5 minutes. Gently aspirate the supernatant and discard it leaving approximately 50 µl of fluid. 9. Resuspend pellet in an appropriate fluid for flow cytometry, e.g. 0.3 mL PBS. The PBS should contain 1% paraformaldehyde (fixative) if samples are not analysed the same day. 10. Analyse on a flow cytometer or store at 2-8 °C in the dark until analysis. Samples can be run up to 24 hours after lysis

    The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene encodes the CD8 alpha chain isoforms CD8, also known as Lyt-2, Ly-2, or T8, consists of disulfide-linked alpha and beta chains that form the alpha(CD8a)/beta(CD8b) heterodimer and alpha/alpha homodimer. CD8a is a 34 kDa protein that belongs to the immunoglobulin family. The CD8 alpha/beta heterodimer is expressed on the surface of most thymocytes and a subset of mature TCR alpha/beta T cells. CD8 expression on mature T cells is non-overlapping with CD4. The CD8 alpha/beta homodimer is expressed on a subset of gamma/epsilon TCR-bearing T cells, NK cells, intestinal intraepithelial lymphocytes, and lymphoid dendritic cells. CD8 is an antigen co-receptor on T cells that interacts with MHC class I on antigen-presenting cells or epithelial cells. CD8 promotes T cell activation through its association with the TCR complex and protein tyrosine kinase lck.
