Protein kinase C ('PKC', EC 2.7.11.13) is a cyclic nucleotide-independent enzyme that phosphorylates serine and threonine residues in many target proteins. It is involved in diverse cellular signaling pathways. Specifically,'PKC'amily members serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the'PKC'amily has a specific expression profile and is believed to play a distinct role in cells'PKC'amily is divided into three groups based on different enzymes' cofactor requirements. They are conventional (c)'PKC'soforms, novel (n)'PKC'soforms, and atypical (a'PKC'soforms respectively. Conventional (c)'PKC'soforms consisting alpha, betaI, betaII and gamma require diacylglycerol (DAG), Ca2+, and phospholipid for activation. Novel (n'PKC'soforms consisting delta, epsilon, eta (also known as PKC-L), Theta and µ(the mouse homolog of human PKCµ known as PKD) require DAG but not Ca2+ for activation. Atypical (a'PKC'soforms, consisting zeta, iota and lambda (the mouse homolog of human'PKC'ota) require neither Ca2+ nor DAG. Other than those listed above, a new'PKC'ember has recently been discovered and is referred to as PKCnu.
